In the process of digestion by the restriction enzymes e.g. EcoRI acts upon the substrate DNA and recognizes the palindromic sequence i.e. GAATTC. It is a site- specific endonuclease and cleaves DNA wherever the GAATTC palindromic sequence is present and produces the sticky ends. The enzymes functions by determining the length of the DNA sequence and once it finds the specific palindromic sequence it binds to the DNA and cuts the 2 strands of double helix at specific positions in the sugar-phosphate backbone of substrate DNA. In this diagram, EcoRI cut the strand of DNA a little away from the centre of palindromic sequence and produces the sticky ends.
